Your hosting service will have to set up the web server to route one request from domaina.com to this directory and domainb.net to another directory. They may give you the option to configure this yourself.

Normally your Hosting Package should include CPanel.. which has a feature that allows you to add domains to your account.. Just make sure you have the DNS pointed to your domains... Your host can supply this information.. If this missed the question your looking for... Try the free route & go with Zone-Edit

One thing I didn't see the other answerers mention, is that it depends on the hosting plan. Most hosting providers sell a hosting account that's attached to one domain.
Lunarpages, for instance, sells both.... but their cheapest plan only allows for one domain, and their 20+/mo. business plan allows for 20 or so.
One of my favorite webhosts for multiple domains is Cornerhost:

Cornerhost allows unlimited domains, as long as you're within your space and bandwidth limits, and also has a very easy control panel to manage them.
If you already have hosting, and don't see any mechanism for selecting or adding new domains... then it probably doesn't allow for it.
